Formal: Excuse me, could you help me with...? / Would you mind helping me? Casual: Hey, can you give me a hand? I need some help with.... Always start polite.

Accepting or refusing help
Accept: Yes, please! Thank you! / That would be great, thanks. / Yes, I could use some help. Refuse politely: No thank you, Ive got it. / Im fine, thanks.
Saying thank you properly
Thank you so much for helping me. / I really appreciate it. / Youve been very helpful. / Thanks a lot! A specific thank you is stronger than a generic thanks.
